- Abstract
We explored the sample collection and DNA extraction method of environmental DNA ( eDNA) teehnique. Droplet digital PCR ( ddPCR) was used to detect the eDNA conteration of the four major Chinese carps in different water layers and different cross-seelions of Y iehang seelion of Yanglze lliver tnmk sln:am. We analyzed the relationship between eDNA eoneenlralion and den-sily of egg and fry, and explored the feasihility of using eDNA teehnique to monilor the nalural n:produelion of the four major Chinese earps. Compared wilh the results from traditional survey methods, the eDNA eoneenlralion of the four major Chinese earps wa.<: exlremely signifieantly eorn:- lated wilh egg and fry densily. 'lbe maximum eoneenlralion wa.-, eonsislenl wilh the peak oeeurrenee time of egg and fry, suggesling thal eDNA teehnique eould prediel the fisherowd behavior for spaw-ning. The investigation of eross-seelion eDNA eoneenlralion during two eonseeulive years sttggested thal the range of fi sh spawning ground wa." 4.5 km upslream to 1 km downsln:am of Yttyangxi. 'lbe pn:dieted spawning ground was loealed within the range estimated by traditional melhods from the lower parl of Carmine Dam to llonghua Tuo. Our n:sults indieale that eDNA teehnique, as a new eeologieal in vestigation method, eould more aeeuralel y determine the distribution of fish with a certain population size and thus wilth a good application prospect.
