Restriction fragment length polymorphism (RFLP) analysis and GISH (genomic in situ hybridization) were performed on euploid plants derived from crosses between Oryza sativa (2n = 24, AA) and two brown planthopper-resistant accessions of O. eichingeri (2n = 24, CC). After screening with 164 RFLP markers, 60 of the 67 euploid plants were identified as introgression lines, each carrying 1–6 small O. eichingeri segments integrated on chromosomes 1, 2, 6, or 10. In the somatic chromosome preparations of F[sub 1] hybrid, O. eichingeri chromosomes, fluorescing greenish-yellow in the sequential GISH, appeared to be longer and to contain more heterochromatin than O. sativa ones, and this karyotypic polymorphism can be used to detect some introgressed O. eichingeri segments in euploid plants. In addition, GISH identification presented direct evidence for the transfer of small segments from O. eichingeri to O. sativa chromosome(s) which were subsequently recognized according to their condensation pattern, arm ratio, and chromosome length. The present results would contribute to the molecular mapping and selection of O. eichingeri - derived brown planthopper-resistant gene and positive yield QTLs.Key words: Oryza sativa, Oryza eichingeri, introgression lines, RFLP, genomic in situ hybridization (GISH).Des analyses RFLP (polymorphisme de longueur des fragments de restriction) et GISH (hybridation génomique in situ) ont été réalisées sur des plantes euploïdes issues de croisements entre l'Oryzasativa (2n = 24, AA) et deux accessions de l'O. eichingeri (2n = 24, CC) qui sont résistantes à la cicadelle brune. Après l'examen de 164 marqueurs RFLP, 60 de 67 plantes euploïdes ont été identifiées comme ayant des introgressions, chacune ayant entre un et six segments chromosomiques de l'O. eichingeri intégrés dans les chromosomes 1, 2, 6 ou 10. Dans des préparations de chromosomes somatiques provenant de l'hybride F[sub 1] , les chromosomes de l'O. eichingeri émettaient une fluorescence vert jaunâtre en GISH séquentielle, semblaient être plus longs et contenir plus d'hétérochromatine que ceux de l'O. sativa. Ce polymorphisme caryotypique peut servir à détecter certains segments introgressés de l'O. eichingeri chez les plantes euploïdes. De plus la détection de segments chromosomiques par la technique GISH a fourni des preuves directes du transfert de petits segments de l'O. eichingeri dans les chromosomes de l'O. sativa. Par la suite, ces segments ont été identifiés grâce à leur état de condensation, au ratio de longueur de leurs bras et à la longueur des chromosomes. Ces résultats peuvent contribuer à la cartographie moléculaire et à la sélection du gène de résistance à la cicadelle brune ou de QTL positifs pour le rendement contribués par l'O. eichingeri.Mots clés : Oryza sativa, Oryza eichingeri, lignées d'introgression, RFLP, hybridation génomique in situ (GISH).[Traduit par la Rédaction]
