- Abstract
[Objectives]This experiment was conducted to study the effects of intrauterine growth retardation(IUGR)on proliferation, apoptosis and antioxidant genes and protein expression of porcine placenta. [Methods]In this experiment, the placentas of normal birth weight and IUGR piglets were selected from newly delivered sows, and they were divided into the control group and IUGR group. Each group contained five replicates. Immunohistochemistry(IHC)was used to detect the PCNA expression in the placental cells. The apoptosis of placental cells was detected by TUNEL. RT-qPCR was applied to detect the mRNA expression of NRF2, HO-1, NQO1, SOD1, SOD2, GCLC and GCLM. Western blot(WB)was applied to determine the protein expression of NRF2 and HO-1. [Results]The results showed as follows:Stronger PCNA immunostaining was observed in the control group, while weaker PCNA staining was observed in the IUGR group. The percentage of apoptotic cells was 12.56% in the control group, whereas that of the IUGR group was 19.63%, the apoptosis rate of the IUGR group was significantly higher than that of the control group(P<0.05). The results of RT-PCR showed that compared with the control group, the mRNA expression of NRF2, NQO1, SOD1, SOD2, GCLC and GCLM in placenta of the IUGR group was significantly decreased(P<0.05). While the expression of HO-1 mRNA showed no significant difference between the control group and the IUGR group(P>0.05). The results of WB showed that the protein expression levels of NRF2 and HO-1 in the control group and the IUGR group were not significantly different(P>0.05). [Conclusions]Intrauterine growth retardation could inhibit the proliferation of porcine placental cells, increase apoptosis and decrease the expression of antioxidant genes and proteins in porcine placenta.
