- Abstract
Verticillium dahliae (Kleb), one of the important diseases that causes significant yield losses wherever the cotton and olive were produced. Because the pathogen is soil-borne which makes the management of the disease difficult. Therefore, biological control in fungal diseases is becoming more essential, one of them is use the mycovirus dsRNAs. Three studies on Verticillium dahliae mycoviral dsRNAs have been carried out so far, two of them in cotton and one in olives. In Turkey, no studies were found on the presence of mycoviral dsRNA in V. dahliae, which causes cotton wilting. In this study, the presence of mycovirus dsRNA in 72 V. dahliae isolates obtained from the cotton fields of Aydn/Söke and Koçarlı districts was investigated method. In 12 V. dahliae isolates dsRNA was determined. The specific primers and random hexamer primer designed for the mycovirus of V. dahliae (VdPV1-ol) in olive were used in RT-PCR identification of this determined dsRNA. These primers have been reported to have identical genome sequences with V. dahliae micovirus (VdPV1) in cotton. Furthermore, V. dahliae isolates containing dsRNA did not respond to the apple pathogenicity test.
