- Abstract
The BLAST was applied to Flanzmulina velutipes genomic and transcriptomic data to identify a small Ran-type GTPase protein which we have designated FvRanl. Bioinformatic analysis indicated that the FvRanl gene consisted of 1337 by with a 1079 by coding region, and contained seven exons and six introns. Among the latter, introns 1, 2, 3 and 6 exhibited characteristics compatible with alternative splicing during mRNA processing. Protein homology and phylogenetic analyses revealed FvRanl to contain five G-Box conserved domains and two effector switches, and to have a close genetic relationship with Ran proteins from the basidiomycetes Moniliophthora roreri and Trametes versicolor. RT-qPCR revealed that FvRanl expression levels were significantly higher in stipe tissue at the maturation stage and in the elongation area located 0. 6-1.5 cm from the top of the stipe. FvRanl expression levels in this elongation area were higher when the cultivation bags were placed horizontally on the platform rotating through 360 degrees at a speed of 30 r/min compared to static bags placed in an upright (vertical) position or laid 30 degrees to the horizontal.
