- Abstract
Equid herpesvirus 2 (EHV-2), one of the important pathogens causing abortions in horses, seriously endangers the healthy development of the horse industry. To investigate whether EHV-2 is the cause of abortion in Yili horses, specific PCR was used to detect EHV-2 in 93 aborted fetus lung tissues of Yili horses collected from six large-scale breeding farms and surrounding retail households in the Zhaosu country. PCR results showed that the EHV- 2 glycoprotein B (gB) gene was detected in three aborted samples, with a positive rate of 3.2% (3/93). Sequence analysis revealed that the EHV- 2 gB genes from China shared 99.9%- 100% nucleotide sequence identity and 99.6%- 100% amino acid sequence. In contrast, the EHV- 2 gB gene from China shared 91.6%-96.4% nucleotide sequence identity and 90.8%-96.2% amino acid sequence identity with the EHV-2 reference strains from UK, Switzerland, Brazil, Germany, Australia, Poland, and Iceland. In addition, a comparison of the amino acid sequences in gB protein revealed 12 different amino acid differences of the three EHV-2 detected in this study from the EHV-2 reference strains Cj-31-06 and Cj-07 reported in Xinjiang. In comparison, 26 amino acid differences exist from other reference strains. These results showed the genetic diversity of Chinese EHV- 2 gB genes. The phylogenetic tree based on the gB gene sequences revealed that Chinese EHV- 2 were closely related and clustered together with EHV- 2 reference strains from Poland, UK and Switzerland, but diverged from the lineages of Chinese EHV-2 reference strains from horse respiratory diseases. In this study, we firstly reported the detection and genetic evolution analysis of the EHV- 2 gB genes from the aborted samples of horses from China. These findings provide fundamental data for the prevention, control and epidemiological study of EHV-2 in China.
