- Abstract
In weakly acidic Britton-Robinson buffer solution, chitosan (CTS) reduced the molecular absorption of direct blue 86. With the increase of CTS concentration, the absorbance of the system decreased gradually, and the fading effect of CTS on the detection system was further enhanced after Tween-20 was added to this system. The absorption spectrum of this system was scanned, with absorbance without CTS recorded as A0 and absorbance with CTS measured as A, and ΔA=A0-A was calculated. At the maximum absorption wavelength of 668 nm and CTS concentration of 0.050 ~ 5.60 µg/mL, the absorbance difference ΔA presented a good linear relationship with the mass concentration of CTS. The regression equation of the detection system was ΔA = 0.0931c - 0.0102 (c : µg/mL), with R² = 0.9971 and detection limit of 0.053 µg/mL. Therefore, a new fading spectrophotometric method for the quantitative determination of CTS based on CTS-direct blue 86-Tween-20 was established. It had the advantages of simplicity, rapidity, high stability and strong anti-interference ability. It has been applied to the quantitative analysis of chitosan in health food with satisfactory results.
