Such changes may weaken the interaction between the -sheet structure and helix structure in the wild-type protein and causes that the mutant protein structure presents an unstable state. Overexpression of IL-36Ra from set without myc-tag in HEK293 cells and detecting with anti-IL-36Ra antibody by Western blotting shows that both C59M and C80M have much lower protein level than WT (Fig. This could explain why the protein expression, not mRNA level, of mutant IL-36Ra at p.Tyr20Cys is much lower than that of nonmutant IL-36Ra in cells transfected with the corresponding constructs.
