- Abstract
This article discusses the importance of accurately estimating fungal biomass in order to understand the carbon cycle in terrestrial ecosystems. The authors introduce a new liquid chromatography-mass spectrometry (LC-MS) method for measuring ergosterol concentration in soil samples, comparing it to the traditional high-performance liquid chromatography-ultraviolet (HPLC-UV) method. The study conducted in Finland found that the LC-MS method produced more reliable results with higher precision. The authors also propose an extraction protocol using KOH in methanol with cyclohexane for liquid-liquid extraction. They emphasize the need for harmonization of methodologies and highlight the lower detection limit of the LC-MS method compared to HPLC-UV.
