- Abstract
배경 The huge threats to increase the nosocomial infections associated with Gram negative bacteria (GNB) and the spread of multi-drug resistant GNB pose world health problem. Pseudomonas aeruginosa (P. aeruginosa) as a common opportunistic pathogen among GNBs can cause healthcare associated infections. However, P. aeruginosa also produce the outer membrane vesicles (OMVs) contained several virulence factors for antibacterial activity. This study investigates the antibacterial activity of P. aeruginosa derived OMVs against GNB strains. 방법 Four clinical P. aeruginosa strains were isolated, and one standard P. aeruginosa strain was evaluated. All strains were tested for their antibacterial activity against other GNB species by cross streak assay. Among them, concentrated OMVs was extracted by selecting clinical P. aeruginosa which showed strong antibacterial effect against Acinetobacter baumannii (A. baumannii), and standard P. aeruginosa as control. The OMV extraction of P. aeruginosa was performed by high speed centrifuge, tangential flow filtration system, and ultracentrifugation. The quantification, Sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis (PAGE), and microscopic visualization by transmission electron microscope (TEM) were conducted for confirming the extracted OMVs. The antibacterial activity of P. aeruginosa derived OMVs was tested using disk diffusion method. 결과 It was found that only one P. aeruginosa cultured from urine isolate shown extensive growth, and in- hibited all tested A. buamannii in cross streak assay. The P. aeruginosa strain inhibiting all A. baumannii shown the about four-fold higher quantity of protein (1665μg/ml), different band pattern (10 - 30kDa, 50kDa) in SDS-PAGE, and variable OMV sizes (20 - 498nm) in TEM compared to OMVs of standard P. aeruginosa. However, there was not shown the inhibitory activity of the P. aeruginosa derived OMVs (5, 10, 20, 40, and 50 ug/ml) against A. baumannii in the disk diffusion method. 결론 The different protein profiles of OMVs between P. aeruginosa species might be correlated the clinical phenomenon. However, antibacterial activity of single cultured P. aeruginosa derived OMVs could be suggested the possibility that inhibition pathway might involve proteins that are expressed only in other condition. Therefore, it would be needed further research for evaluate the additional mechanisms of the antibacterial function of OMVs.
