Abstract
The author reflects on the development of the methods for targeted genome editing. It contends that zinc-finger nucleases (ZFNs) were shown to cleave site specifically in cells, while transcription activator-like effector nucleases (TALENs) consist mainly of proof-of-principle reports. It argues that targeted nuclease can be used to make mutations and change of introduce sequences into endogenous genes in cultured cells.
