- Abstract
Twenty one samples of Morchella spp., including three cultivated M. importuna samples, five M. eximia samples, ten M. sextelata samples and three wild M. elata samples were subjected to tissue isolation to obtain 348 isolates. These isolates were then analyzed for their mating type gene makers respectively. The results showed that the detection rate of the two representative mating type markers Mat1-1-1 and Mat1-2-1 were similar (Mat1-1-1:Mat1-2-1=267:278). There was only one kind of mating genotype detected in 153 isolates, accounting for 43.96% of the total number of the tissue isolates. For these 153 isolates with the other mating genotype missing, the probability of mating gene deletion for Mat1-1-1 and Mat1-2-1 were similar, with 83 isolates missing Mat1-1-1 (23.85%) and 70 isolates missing Mat1-2-1 (20.11%), respectively. For M. septemtelata 19-37 and M. septemtelata 19-43, only Mat1-2-1 was detected. In contrast, M. septemtelata 19-44 only contained Mat1-1-1. Initial correlation analysis between colony morphology of tissue isolates and the mating type gene deletion phenomenon indicated that isolates with slow germination and colonies of sparse hyphae were more likely to have a deletion of either mating type gene than isolates with fast germination and dense colonies.
