- Abstract
The present study aimed to compare the efficiency between culture and molecular (multiplex PCR) methods in the identification of gram negative bacteria isolated from necrotic root canals of patients with periodontitis. periodontics is the infection of the root canal of the tooth and is the primary etiologic agent of the different forms of perpendicular inflammatory diseases . The pathological process is started when the dental pulp becomes necrotic, usually as a sequel to caries, and then infected by micro-organisms that are usually normal inhabitants of the oral cavity. The principle of study was conducted on 100 patients with Periodontics collected from private clinics in Baghdad.Sampling was performed by a sterile swap. The specimens were primarily evaluated, as per standard methods. Bacterial samples were evaluated by a microbiological technique specific for gram negative bacteria, used for isolation and identification of sampled strains.Then the bacterial specimens undergo for DNA extraction and molecular detection for 16S rDNA gene by Multiplex PCR.The predominant species that have been identified in present study were K. pneumonia ‚ P.aeruginosa and E.coli . The prevalence rate with bacterial culture and biochemical methods was 24.2%‚ 18.2% and 27.3%, respectively. While the prevalence rate with multiplex PCR was (30%‚ 30% and 40%) respectively. The difference between the culture and PCR methods in identification was statistically significant (P=0.8). The study concludes that The Multiplex PCR is more sensitive and specific for the detection of bacterial pathogens than the culture methods.
