Detection of human papillomavirus by PCR genotyping and immunostaining in a population of bosnian women.

- Abstract
Objectives: To determine the prevalence of human papillomavirus (HPV) among Bosnian women. Methods: Cervical swab samples and smears were collected from 97 women from three disease defined groups: Group 1: Patients who had an abnormal Papanicolaou (PAP) cytology report (N= 34); Group 2: Patients who had a history of genitourinary infections (N=22), Group 3: Patients not in either group 1 or 2 (N=41). Age groups were defined as: 20-24 yr, 25-29 yr, 30-34, 35-39 yr, 40-44 yr, 45-49 yr, > 50 yr. Specimens were collected from December 2004 to January 2005 at two sites in Sarajevo. Specimens were shipped to Johns Hopkins University for testing. Swab samples were tested for 37 high risk (HR) and low risk (LR) HPV types by PCR (Roche prototype line blot assay) and for HR HPV by the Hybrid capture-2 (HC2) test (Digene). Slide samples were stained for p-16 protein (Ventana). Results: The overall prevalence for high risk HPV by the HC2 test was 22.7% (22/97). The highest HR-HPV DNA prevalence by HC2 was 31.8% (7/22) in women age 25-29 years. The lowest prevalence was 4.5% (1/22) in women age 20-24 years. Combined HR-HPV and LR-HPV prevalence by Roche PCR was 29.9 % (29/97). The highest prevalence 34.5% (10/29) was in women age 25-29 years. The prevalence of positive cytoplasmic immunostaining by p16 antibodies was 9.3% (9/31) of total HPV positive patients (HC2 and PCR positives all together). The most prevalent HPV genotypes were: HPV 16 (9/29), HPV 52 (4/29), HPV 31, 53, and 67 (3/29). Conclusion: High-risk HPV infection was prevalent among Bosnian women. These results demonstrated that HPV DNA testing was a useful indicator for prevalent of HPV infections and could be adjunct test to PAP testing as a part of regular cervical screening programs among women in Bosnia and Herzegovina in the future.
