- Abstract
The article discusses developments in adrenocortical toxicology and provide a standardized experimental strategy for the investigation of toxicological effects on adrenocortical function and mechanism elucidation. The human adrenal conditions are reviewed and adrenocortical suppression is identified as the toxicological hazard of most concern. The proposed strategy involves an in vivo rodent adrenal competency test based on adrenocorticotropic hormone (ACTH) challenge to examine specifically adrenocortical suppression. It also highlights the consequences of inhibition of key steroidogenic enzymes and the possible toxicological modulation of other adrenal conditions.
