- Abstract
Introduction: Avulsion is a complex injury that includes total displacement of the tooth from its socket. The most common treatment option for this injury is replantation. However, the success of replantation depends on periodontal ligament (PDL) cell viability and extra oral storage time. Various storage media have been evaluated to maintain PDL cell viability. Objective: To evaluate the effectiveness of amniotic fluid (AF) (amniocentesis/full-term), and Cornisol in maintaining PDL cell viability and apoptosis rate of PDL fibroblasts in comparison with HBSS. Methodology: Periodontal fibroblasts, procured from roots of healthy premolars cultured in a-MEM, were treated with 100% AF (amniocentesis), 100% AF (fullterm), Cornisol, and HBSS. Culture plates were incubated with experimental media at 37°C for 1, 3, 24 h, and 48 h. Assessment of cell viability was made using MTT assay. To assess the rates of apoptosis, cells were fluorescently stained with acridine orange/ethidium bromide or stained with propidium iodide and analyzed by flow cytometry after 24 h of incubation. The effects of the test storage media at each period were evaluated using one-way analysis of variance (ANOVA) test and multivariant ANOVA test, and this was followed by post hoc Tukey's multiple comparison test (P < 0.05). Results: 100% AF (amniocentesis) displayed maximum %cell viability in all four time periods (P = 0.001), followed by 100% AF (fullterm) as compared to HBSS. Cornisol displayed minimum %cell viability at all intervals. Conclusion: Amniotic fluid as a potential storage media for the avulsed tooth can be explored in the future.
