- Abstract
Objective To explore the influence of different solvents extracted from lysimachia on cytotoxicity of human gastric cancer cell line MGC-803 and to determine the content of luteolin. Methods Extracts of different solvents (water, ethyl alcohol and ether) on cytotoxicity of human gastric cancer cell line MGC-803 was detected by u-sing four methyl pyrazole (MTT) salt and sulforhodaminc B (SRB)colorimetric method; the paclitaxel group was used as the positive control. The content of luteolin in different extracts was detected by establishing high performance liquid chromatography (HPLC) method. Results Results of MTT and SRB methods showed that the extracts groups and paclitaxel group expressed certain toxicity on MGC-803 cell. Cytotoxicity of each group (compared by IC50) :taxol group > ether extract group > ethanol extract group > water extract group, and cell toxicity of ether extract group and ethanol extract group was significantly higher than that of water extract group (P<0.05). Luteolin in 0.03 ~0.77 mg/ mL showed good linear relationship (r = 0.999 9, n =7), with good precision (RSD=0.17%, n =6), good reproducibility (RSD=0.32%, n =6), good recovery rate of samples (mean value 101. 13%, RSD = 1.43%, n =6), and good stability (RSD = 0.16%, n =6). The content of luteolin: ether extract (0.98%)> ethanol extract (0.65%)> water extract (0.43%). Conclusion Luteolin has certain anti-cancer activity, which may be related to the content of luteolin and provides theorectical evidence for its clinical application.
