- Abstract
The development of nucleic acid release systems to target cells for therapeutic purposes faces important challenges in the efforts to overcome extracellular and intracellular barriers and achieve greater efficiency in transfection. A better knowledge of the molecular mechanisms that mediate this complex process is inescapable. In this review we provide an overview of several systems or methods for non-viral nucleic acid release and the strategies developed in them to overcome the different barriers. The endosomal compartment is considered one of the crucial bottlenecks to be overcome by these systems. We analyze some of the hypotheses which describe how different elements that are included in these systems mediate the endosomal escape.
