- Abstract
A simple and effective method was established for separation and characterization of flavonoid constituents in Radix Astragali (RA) by combinationof ultra-high-pressure liquid chromatography with LTQ-Orbitraptandem mass spectrometry (u-HPLC-LTQ-Orbitrap-MSn). For three majorstructural types of flavonoids, the proposed fragmentation pathways andmajor diagnostic fragment ions of isoflavones, pterocarpans and isoflavanswere investigated to trace isoflavonoid derivatives in crude plantextracts. Based on the systematic identification strategy, 48 constituentswere rapidly detected and characterized or tentatively identified, many ofwhich were first reported in RA. The u-PHLC-LTQ-Orbitrap MSn platformwas proved as an effective tool for rapid qualitative analysis of secondarymetabolite productions from natural resources.
