Loss of Y Chromosome in Bone Marrow Cells: Age or Disease Related?

- Abstract
Controversy exists over the relevance of Y chromosome loss in metaphase cells from bone marrow samples as to whether it is related to advanced age or hematological disease. To study this, 72 bone marrow samples from males 50 years or older, with or without hematological disease, were analyzed using both conventional karyotyping and XY fluorescence in situ hybridization (FISH). Similar results for Y chromosome loss suggested that the karyotype evaluation identified Y chromosome loss as readily as FISH. The study was then expanded to evaluate archival cytogenetic and pathologic data on 671 bone marrow samples from males 50 years of age or older over a period of 6 1/2 years. Loss of the Y chromosome in male patients with disease (7.5%) was not significantly different from loss of the Y chromosome in bone marrow cells from males with no disease (9.7%). Our results suggest that loss of the Y chromosome may be more related to patient age than disease.
