- Abstract
Obesity is major risk factor for insulin resistance, type 2 diabetes and cardiovascular disease. Inhibition of adipose tissue differentiation represents a major issue to develop a comprehensive strategy to prevent and treat obesity. β-glucan from Chaga mushroom (Inonotus obliquus) is plant hemicellulose polysaccharide (soluble fiber) that is recognized as hypocholesterolemic and hypoglycemic compound. The role of β-glucan from Inonotus obliquus in adipocyte differentiation was not evaluated, therefore we examined the effects of β-glucan from Inonotus obliquus on adipocyte differentiation in 3T3L1 cells. To examine the effect of β-glucan on adipocyte differentiation in 3T3L1 cells, 3T3L1 cells were differentiated in the presence or absence of β-glucan and expression of various adipocyte-specific transcription factors were examined. β-glucan inhibited adipogenesis in 3T3L1 cells by downregulating several adipocyte-specific transcription foctors including peroxisome proliferators-activated receptor-Γ C/EBP-α, which are critical for adipogenesis in vitro. We had also found that adipose fatty acid-binding protein (aP2) and lipoprotein lipase (LPL) that were involved in adipocyte differentiation and adipocyte specific genes, were suppressed by β-glucan. This study suggests that β-glucan from Inonotus obliquus inhibits adipocyte differentiation via downregulation of expression of adipogenic factors in 3T3L1 cells. Our data suggest the potential value of β-glucan from Inonotus obliquus as anti-obesity agent.
