- Abstract
The method used for the isolation and estimation of porphyrin α of heart muscle has been applied to bovine tissues as well as to bakers' yeast and <em>E. coli</em>. Only from skeletal muscle was nearly pure porphyrin α obtained in good yield; further fractionation yielded pure porphyrin α in small yield. Pure porphyrin α was also obtained from liver, in very small yield. The porphyrin α from kidney, liver and cow udder differed from that of heart in its absorption spectrum (low ratios of bands III to band IV), probably because it is altered during the extraction. Only a very small fraction of porphyrin α, highly impure, was obtained from yeast and <em>E. coli</em>. Evidence has been found in kidney for the presence of a protoporphyrin ester which is not extracted from ether by 8 p.c. HCl, but which can be converted to protoporphyrin by strong HCl. Protoporphyrin-like compounds also accompany porphyrin α in the extracts from the microorganism, but these were not converted to protoporphyrin by saponification with acid.
