Elastase Inhibitory Activity of Extracts from Polygonum cuspidatum.

- Abstract
The elastase inhibitory activity of extracts from Polygonum cuspidatum was studied in this paper. The crude extract 1 (CE1) was extracted with hot water from P. cuspidatum. The crude extract 2 (CE2) was separated from CE1 by using polyamide column chromatography. Extracts were identified by qualitative and quantitative analysis. The elastase inhibitory activity of extracts was studied by ultraviolet spectrum, and fluorescent spectrometry was used to investigate the interaction of elastase with the CE1, CE2 and polydatin. The results indicated the polydatin was the main component in CE1 and CE2, the content of polydatin were 41.01% and 69.59%. CE1, CE2 and polydatin all had significant inhibitory effect on elastase. When the concentration was 2.0 mg/mL, the inhibiting ratio of CE1, CE2 and polydatin were 53.56%, 61.27% and 82.53%, respectively. Besides, these compounds had apparent endogenous fluorescence quenching effects. When the concentration was 0.1 mg/mL, the fluorescence-quenching ratio of CE1, CE2 and polydatin were 70.38%, 72.90% and 75.99%, respectively.
