- Abstract
A translocação bacteriana é o movimento de bactérias através da parede intestinal, sendo o abdômen agudo por lesões intestinais um dos fatores predisponentes. A confirmação da translocação bacteriana para líquido peritoneal, em equinos, tem sido limitada por técnicas baseadas em cultura. Objetivou-se utilizar o sequenciamento de última geração para detectar DNA bacteriano no líquido peritoneal de equinos com cólica. Foram utilizados 20 cavalos adultos submetidos à laparotomia exploratória por lesões de intestino grosso (grupo cólica) e 4 cavalos saudáveis (grupo controle). Realizou-se coleta de líquido peritoneal, sob anestesia geral inalatória, imediatamente após a incisão na linha média para o grupo cólica e em estação para os do grupo controle. Também foram utilizados dois controles negativos (grupo Qiagen), possuindo solução do kit de extração de DNA Qiagen, no intuito de excluir contaminação ambiental. A região V4 do gene 16S rRNA foi sequenciada com Illumina MiSeq. A análise de variância molecular foi utilizada para comparar as comunidades bacterianas. As amostras também foram submetidas aos métodos de cultura padrão. DNA bacteriano foi encontrado em todas as amostras, enquanto o método de cultura padrão detectou bactérias em apenas uma amostra. A composição bacteriana no líquido peritoneal do grupo cólica foi significativamente diferente no grupo controle (p = 0,001) e 80% dos equinos com cólica apresentaram composição bacteriana similar, que foi representada principalmente por Gemella, Actinobacillus e Bacterioides spp. Todas as demais amostras apresentaram composição semelhante aos controles negativos, indicando ausência de bactérias. Não houve diferença na composição bacteriana das amostras quanto ao tempo de evolução, tipo de lesão (deslocamento, obstrução intraluminal ou torção) ou sobrevida. Conclui-se que o sequenciamento de última geração é mais sensível do que a cultura para detectar bactérias no líquido peritoneal de cavalos com cólica. Métodos moleculares podem ser usados para direcionar táxons comuns presentes durante a translocação bacteriana.
