- Abstract
The inflammatory response is a marvel of cellular orchestration and timing, often involving the sequential recruitment of both innate and adaptive elements of the immune system. One of its earliest initiating events can occur through activation of mast cells, leading to the immediate release of biologically active molecules that alter the local tissue environment, and rapidly thereafter to the production of secondary mediators that promote cellular infiltration. Leukotrienes are lipid-based messengers of this latter class, able to prolong and extend inflammatory responses by increasing vascular permeability and recruiting myeloid leukocytes. This study demonstrates an unexpected function for the leukotriene LBT4 as a chemoattractant for activated CD4+ and CD8+ T cells expressing BLT1, the high-affinity LTB4 receptor. These findings establish the LTB4-BLT1 pathway as providing an essential early link between the innate and adaptive effector responses to inflammation, and offer new possibilities for therapeutic intervention in the allergic and chronic inflammatory states.
