Synthesis and processing of bone sialoproteins during de novo bone formation in vitro.

- Abstract
Bone sialoprotein (BSP) and osteopontin (OPN) are sulphated and phosphorylated sialoglycoproteins that regulate the formation of hydroxyapatite crystals during de novo bone formation. To gain insights into the relationship between the synthesis and posttranslational modification of BSP and OPN and the mineralization of bone, pulse–chase studies were conducted on cultures of newly forming bone nodules produced by fetal rat calvarial cells in vitro. Cultures were pulse labelled with [sup 35] SO[sub 4] , or with either [sup 32] PO[sub 4] or [γ-[sup 32] P]ATP to study intracellular and extracellular phosphorylation, respectively, and chased in isotope-free medium for various times up to 24 h. The presence of radiolabelled BSP and OPN was determined in the cells, in culture medium, and in various tissue compartments obtained by dissociative extraction with 4 M GuHCl (G1), 0.5 M EDTA (E), and again with 4 M GuHCl (G2) and a bacterial collagenase digestion of the demineralized collagenous tissue residue. With each isotope employed, radiolabelled BSP and OPN were detected in the E extract within the 1-h chase period and increased in amount with time. Similarly, [sup 35] SO[sub 4] - and [sup 32] PO[sub 4] -labelled BSP increased in the G2 extract, but OPN was not detected. In the G1 extract the [sup 35] SO[sub 4] -labelled BSP decreased with chase time, whereas the [sup 32] PO[sub 4] -labelled BSP increased. No differences were evident in the profiles of BSP labelled with [sup 32] PO[sub 4] or [γ-[sup 32] P]ATP. In the absence of β-glycerophosphate, which is required for optimal mineralization of the bone nodules, [sup 35] SO[sub 4] -labelled BSP was increased in the medium and G1 extract and decreased in the E extract and G2 extract after 3 h. In addition to differences in the tissue compartmentalization of BSP and OPN, these studies indicate that [sup 35] SO[sub 4] is lost from BSP during mineralization and that isoforms of BSP exist with a selective affinity for the organic and mineral phases. Moreover, the additional phosphorylation of BSP and OPN catalyzed by ectokinase activity does not appear to alter the distribution of these sialoproteins.Key words: biosynthesis, bone, bone sialoprotein, osteopontin, mineralization, posttranslational modification.La sialoprotéine osseuse (BSP) et l'ostéopontine (OPN) sont des sialoglycoprotéines sulfatées et phosphorylées qui règlent la formation des cristaux d'hydroxyapatite durant la formation de l'os de novo. Afin de mieux comprendre la relation entre la synthèse et la modification post-traductionnelle de la BSP et de l'OPN et la minéralisation de la substance osseuse, des études de marquage court suivi d'une chasse ont été faites en utilisant des cultures de nodules osseux se formant à partir de cellules de boîte crânienne de foetus de rat. Les cultures ont été marquées brièvement avec du [sup 35] SO[sub 4] , ou avec du [sup 32] PO[sub 4] ou du [γ-[sup 32] P]-ATP afin d'étudier la phosphorylation intracellulaire et extracellulaire, respectivement, puis l'incubation a été poursuivie dans un milieu sans isotope pendant 24 h. La présence de la BSP et de l'OPN marquées a été déterminée dans les cellules, dans le milieu de culture et dans divers compartiments tissulaires après extraction avec du chlorhydrate de guanidine (GuHCL) 4 M (G1), puis, avec de l'EDTA 0,5 M (E) et, encore une fois, avec du GuHCL 4 M (G2), et après digestion du tissu collagénique résiduel déminéralisé par une collagénase bactérienne. Dans l'extrait E, la BSP et l'OPN marquées avec chacun des isotopes sont détectées pendant la première heure de chasse et leur quantité augmente en fonction du temps. Dans l'extrait G2, la BSP marquée avec du [sup 35] SO[sub 4] ou avec du [sup 32] PO[sub 4] augmente de la même façon, mais l'OPN n'est pas détectée. Dans l'extrait G1, la BSP marquée avec du [sup 35] SO[sub 4] diminue avec le temps lors de la chasse, alors que la BSP marquée avec du [sup 32] PO[sub 4] augmente. Il n'y a pas de différence évidente dans les profils de marquage de la BSP avec du [sup 32] PO[sub 4] ou du [γ-[sup 32] P]-ATP. En absence de β-glycérophosphate, qui est essentiel pour une minéralisation optimale des nodules osseux, la BSP marquée avec du [sup 35] SO[sub 4] augmente dans le milieu et dans l'extrait G1, mais elle diminue dans les extraits E et G2 après 3 h. En plus des différences de compartimentation tissulaire de la BSP et de l'OPN, ces études montrent que le [sup 35] SO[sub 4] est éliminé de la BSP au cours de la minéralisation et qu'il y a des isoformes de la BSP ayant une affinité sélective pour la phase organique et la phase minérale.
