- Abstract
[Objective] Genome-wide identification of OSCA family members (TaOSCAs) was conducted to clarify the potential biological functions of TaOSCA in wheat.[Method] The TaOSCA members in wheat were identified by BLASTP combined with the screening of conserved domains. Phylogenetic, gene structure and transmembrane domain of TaOSCA gene family members were analyzed by bioinformatics methods. The expression profiles of TaOSCA in different organs/tissues of wheat at different developmental stages were analyzed using published transcrip tome data. PEG-simulated osmotic stress treatment was performed using Chinese spring wheats, and the expression patterns of TaOSCAsin leaves under osmotic stress were detected by real-time PCR (RT-qPCR).[Result]A total of 35 TaOSCAs were identified in wheat, and members of the TaOSCA family were divided into 4 subfamilies of TaOSCA I, TaOSCAR, TaOSCAHI and TaOSCAIV, with 15,16, 3 and 1 members, respectively. The gene structure analysis showed that TaOSCA genes contained multiple exons except TaOSCA4. 1-4B. The transmembrane domain analysis showed that TaOSCA proteins contained 8~11 transmembrane domains (TMs) except for TaOSCA2. 3-4B with 5 TMs. The expression profiles of TaOSCAs in different stages and tissues of wheat showed that TaOSCA 1. l-3B/3D, TaOSCAl. 2-1A/1B/W, TaOSCAl. 3-lA, TaOSCAl. 6-1A/1D and TaOSCA3. 1-2B/2D were highly expressed in the whole growth stage, TaOSCA 1. 4-2A/2B/2D were highly expressed in the root tissue in different stages, TaOSCA 1. 5-1B/1D were highly expressed in the panicle during the reproductive growth stage, and CTaOSCAl. 6-1B and TaOSCA2. 6-3B/3D were highly expressed in grains during reproductive growth. These genes may play important functions in root, spike and grain development. The RT-qPCR detection of leaves under osmotic stress treatment showed that TaOSCA2. 4 and TaOSCA4. 1 expressions were inhibited under osmotic stress, TaOSCA2. 5 expression was insignificantly changed, while expressions of 11 TaOSCAs including TAOSCA 1. 1 and TaOSCAl. 2 were up-regulated under osmotic stress. It is indicated that these genes may play a role in response of wheat seedlings to osmotic stress.[Conclusion] TaOSCA gene may play an important role in responses of wheat to osmotic stress.
