- Abstract
Headspace solid-phase microextraction and gas chromatography-mass spectrometry were used to compare the effects and characteristics of different coated fiber extraction heads and different chromatographic columns on the extraction and separation of volatile components in the water extract of Allium tenuissimum flowers. Combined with the laboratory's preliminary test results, a total of 133 types of volatile substances were detected, including 10 types of sulfur, 22 types of esters, 8 types of acids, 46 types of hydrocarbons, 15 types of alcohols, 8 types of ketones, 10 aldehydes, 8 phenols, 2 ethers and 4 other compounds. Among them, the key aroma components of the water extract of Allium tenuissimum flowers included p-vinyl guaiacol, 1, 3-dithiane, dimethyl trisulfide, dimethyl disulfide, methyl propyl disulfide, 2, 5-dimethylthiophene, 3, 4-dimethylthiophene and other substances. According to the total ion chromatogram, the separation effect of the volatile components in the water extract of Allium tenuissimum flowers with HP-5 chromatographic column was better than that with RTX-WAX chromatographic column. At the same time, the differences of the number of components and the total relative content detected by different extraction heads were compared, and the optimal extraction head was selected by principal component analysis, the results showed that, 85μm PA extraction head had better separation and identification effects with HP-5 chromatographic column; 65μmPDMS/DVB extraction head had better separation and identification effects with RTX-WAX chromatographic column.
