Whole genome analysis of protracted outbreak of Salmonella enterica serovar Enteritidis from Republic of Korea, 2020-2022.

- Abstract
배경 Salmonella enterica serovar Enteritidis is a common pathogens of human gastroenteritis in worldwide. From Oct 2020 to Feb 2022, 9 food-poisoning outbreaks caused by S. Enteritidis continuously occurred. As molecular epidemiologic analysis network, PulseNet Korea identified that all 112 isolates from 9 out- breaks have one SEGX01.049 pattern type by pulsed field gel electrophoresis(PFGE). In this study, we investigated genetic relationships of S. Enteritidis (SEGX01.049) by whole genome analysis 방법 We analyzed 27 S. Enteritidis(SEGX01.049) isolates selected from 9 outbreaks and 2 sporadic diarrhea cases. Antimicrobial susceptibility testing was performed using broth microdilution method. Whole genome sequencing (WGS) was performed by Illumina Miseq platform. The genetic relatedness was analyzed by high-quality single nucleotide polymorphism (hqSNP) analysis. 결과 The genome size obtained by performing WGS was approximately between 4.64Mbp-5.51Mbp, and the GC ratio was 52.0%. Only one outbreak had resistance to ampicillin (>32 mg/mL). WGS revealed 3 different cluster: group A (n=21, 77.8%), group B (n=4, 14.8%), and group C (n=2, 7.4%). In particular, hqSNP analyses showed that 21 isolates (cluster A) were closely related with SNP differences (range = 0-25), suggesting that the S. Enteritidis isolates found in outbreak all most shared a common source. 6 isolates (cluster B and C) were determined to not be closely related and were excluded from the same outbreak. 결론 This finding means that the same pathogen is continuously found in various regions of Republic of Korea. A continuous monitoring system is needed to prevent the spread of major pathogenic S. Enteritidis.
