Análisis de la metaciclogénesis de Trypanosoma cruzi en diferentes unidades discretas de tipificación.

- Abstract
Introducción: Trypanosoma cruzi presenta una amplia variabilidad genética; es por ello que esta especie se ha dividido en subgrupos, identificados por marcadores moleculares, genéticos e inmunológicos. Durante el proceso de metaciclogénesis, los epimastigotes no infecciosos se convierten en tripomastigotes metacíclicos infecciosos. Por lo tanto, el objetivo de este estudio fue determinar si se presentan diferencias en el tiempo necesario para alcanzar el punto máximo de metaciclogénesis en cultivos de diferentes unidades discretas de tipificación (DTU). Métodos: Se cultivaron las cepas TcI(DA/MG), TcII(Y), TcIII(845), TcIV(85) y TcVI(Tulahuen), y tres réplicas biológicas en medio LIT con suplemento de suero fetal bovino, propiciando condiciones de estrés nutricional que den lugar a la metaciclogénesis. Resultados: El punto de inicio de la metaciclogénesis para DA se encontró en el día 3, MG (4), TCII (6), TcII, TcIV(3) y TcVI (4). Entre las cepas de la DTU TcI, se encontraron diferencias estadísticamente significativas desde el día 5. Sin embargo, el número de MT fue superior en MG con respecto a DA (52'000.000 MT/9'000.000 MT). Por otra parte, la cepa que presentó una menor cantidad de MT fue TcIV. Los datos siguen una distribución normal y, en el análisis ANOVA de dos vías, se encontraron diferencias estadísticamente significativas con un valor p<0,0001 para la cantidad de tripomastigotes metacíclicos en función de la cepa/día. Conclusión: La cepa que presentó una mayor eficiencia en el proceso de metaciclogénesis, fue la cepa MG, lo cual demuestra que existen diferencias, no solo entre las DTU, sino además, entre cepas de la misma DTU, lo cual podría relacionarse con circulación y transmisión de la misma.
