- Abstract
In order to accurately determine the acid value of Ganoderma lingzhi spore powder products and clarify the key factors affecting acid value, a high performance liquid chromatography-evaporative light scattering detector (HPLC-ELSD) analysis method was established to determine the content of free fatty acids in spore oil. By optimizing the chromatographic conditions, a HPLC quantitative determination method for determining linoleic acid, palmitic acid and oleic acid was established. This method had good precision, repeatability and stability, and could be used for the quantitative determination of linoleic acid, palmitic acid and oleic acid components in G. lingzhi spore powder, spore oil and related products. Compared with the national standard chemical method, the matching degree of the HPLC-ELSD is better, and the method has high sensitivity and can determine the content of different fatty acids. Through comparative analysis, it was found that the storage time had the greatest effect on the fatty acid content of G. lingzhi spore powder, and the producing area and harvest time also affected the size of acid price. The fatty acid content of eight different brands of commercially available G. lingzhi spore oil products showed varied greatly, ranging from (1.90±0.12) mg/g to (99.81±10.21) mg/g. The newly established HPLC-ELSD method can accurately quantify the free fatty acids in G. lingzhi spore powder, which provides a scientific basis for the quality control of G. lingzhi spore products on the market.
