- Abstract
In this study, the osteogenic properties and anti-osteoporosis activity of calcium hydroxyapatite (HA) obtained from Katsuwonus pelamis tuna bone and water-soluble structured forms of HA, such as the structured HA chloride citrate compound (HA-Cl-Ci) and the structured HA hydroxide lactate compound (HA-OH-Lac), were investigated. There was no significant cytotoxicity towards MC3T3-E1 pre-osteoblast and Caco-2 cells. HA enhanced osteogenic proliferation and differentiation after 7, 14, and 21 days of treatment, as demonstrated by alizarin red staining, alkaline phosphatase (ALP) activity, and qRT-PCR. Also, the anti-osteoporosis efficiencies of HA, HA-Cl-Ci, and HA-OH-Lac were evaluated in dexamethasone-induced MC3T3-E1 osteoblasts. HA, HA-Cl-Ci, and HA-OH-Lac had the potential to prevent glucocorticoid-induced osteoporosis effects. In addition, the transcellular transport of these compounds across the Caco-2 cell monolayers was investigated. Ca, P, and Mg derived from HA, HA-Cl-Ci, and HA-OH-Lac were absorbed in the intestinal tract. The absorption of these minerals may have resulted in the osteogenic proliferation, differentiation, and anti-osteoporosis activity.
