- Abstract
The microalgae Dunaliella salina is a valuable resource, especially for ß-carotene production. However, the biomass harvesting is currently an obstacle for its large-scale exploitation, which makes the choice of the harvesting method an essential and decisive step. The aim of this work is to study the efficiency of two harvesting methods, centrifugation and flocculation at different pH and their effects on the nutritional quality of the harvested biomass. For the centrifugation six rotations were tested. The pH values tested for flocculation were 4; 6.5; 8.5; 10; 10.5; 11.5 and 12. The effect of the harvesting method on the biomass quality is estimated by the determination of proteins, carbohydrates and fatty acids profile which is carried out by Gas Chromatography (GC) after transmethylation. Total biomass recovery was obtained by flocculation at a basic pH above 10.5. While a maximum recovery of 94.85 ± 0.47% at 2500 rpm for 10 min was obtained for centrifugation. The highest proteins and carbohydrates amounts were detected in the biomass harvested by centrifugation, 54.51 ± 0.27% DM and 18.13 ± 1.11% DM, respectively. However, the biomass harvested by flocculation has a protein amount four times lower (13.39 ± 0.13% DM) and for the carbohydrates content six times lower (3.02 ± 0.22% DM). The fatty acid contents in the biomass harvested by flocculation were also affected, some have even disappeared. The harvesting method can affect the nutritional quality of the biomass, so it is necessary to choose carefully the method to be adopted.
