- Abstract
Objective To explore the characteristic biomarkers in the serum of sepsis rats. Methods Cecal Ligation and Puncture (CLP) was used to replicate the sepsis rat model, and the rats were divided into 5 groups: group A (sham operation group), group B, group C, group D, group E, each group 10 rats were collected at 0h after operation, group B at 6h after operation, group C at 12h after operation, group D at 24h after operation, and 2mL of heart blood from group E at 48h after operation. After static and centrifugation Collect serum. The metabolomics method based on liquid chromatography-mass spectrometry was used to analyze the changes of serum metabolites in septic rats. Results The cluster analysis of differential ions showed that the differential ions can be found in the serum of group B rats starting at 6 hours after surgery (or even earlier). By 24h, the difference between the ions in the serum of group D and group A was the most obvious and the intensity was the greatest. Principal component analysis (PCA) and partial least squares-discriminant analysis (PLS-DA) found that the metabolites of rats in group A and other groups were different, and the difference in metabolites in group D changed most significantly. Through metabolic pathway analysis, there are 12 characteristic different metabolites between group D and group A, including D-glucosamine 6-phosphate, D-glucosamine phosphate, α-D-glucosamine 1-phosphate, and D-glucosamine 1 -Phosphoric acid, D-galactosamine 6-phosphate, bilirubin, (3Z) -Plant pigment, 15,16-dihydrobiliverdin, 15,16-dihydrobiliverdin IXα, 15,16-dihydro Biliverdin ixa, cis-(Homo) 3-aconitic acid, 2,3-Bis-O-(geranylgeranyl)-sn-glycerol-1-phosphate-L-serine. In D-glucosamine and its phosphorus-containing derivatives, D-glucosamine 6-phosphate, D-glucosamine phosphate, α-D-glucosamine 1-phosphate, D-glucosamine 1-phosphate, D- The content of galactosamine 6-phosphate decreased significantly.
