Transcriptome Analysis Reveals the Effect of Light on Anthocyanin Synthesis in Leaves of Aeonium arboreum 'Halloween'.

- Abstract
Aeonium arboreum 'Halloween' is an indoor foliage succulent plant with coloured photosensitive leaves found mostly in China. However, the current understanding regarding the mechanism of coloured leaf formation in succulents is elusive. Qualitative and quantitative analysis of flavonoids and transcriptome sequencing was carried out on dark and light-treated 'Halloween' leaves to investigate how light affects the leaf colour of the succulents. The findings supported the evidence that the red color of 'Halloween' leaves is mainly attributed to the high accumulation of cyanidin 3-O-galactoside. A total of 89 513 unigenes from the transcriptome were identified, and 65.24% of these have annotations in the NR, NT, KEGG, SwissProt, PFAM, GO and KOG databases. A total of 20 898 DEGs were obtained following the light treatment compared to the dark treatment, including 11804 up-regulated genes and 9094 downregulated genes. The KEGG enrichment analysis identified 20 metabolic pathways with the most significant enrichment of DEGs, including the flavonoid and phenylpropanoid synthesis pathways, which are closely associated with anthocyanin metabolism. Almost all structural genes and transcription factors related to flavonoid metabolism were up-regulated as verified by qRT-PCR, and five transcription factors, AaMYB113, AaMYB114, AaMYB4, AaHY5 and AaCOP1, were initially screened for their possible involvement in regulating light-induced anthocyanin synthesis. This study contributes to a deeper understanding of the effects of light on leaf colouration in Aeonium arboreum 'Halloween' and in succulents.
